Is any configuration required for ProxyClient to accelerate RDP?

There is nothing that needs to be done on the ProxyClient to accelerate RDP traffic.  There will not be a significant gain in performance from ProxyClient unless encryption and compression are disabled on the client.  Even then, there won't be great gains as the client is only seeing the RDP byte cache from its own session and not benefitting from other users RDP to the similar workstations.
